A.? Personal Statement    

I am focus on molecular biology, clinical epidemiology, and evidence-based medicine of thyroid, breast and gastrointestinal cancers. I and my colleagues can perform minimally invasive surgery of hepatobiliary pancreatic diseases. Since 2014 years, I have been the director of Key Laboratory of Hepatobiliary and Pancreatic Disease Prevention and Treatment in Dalian and Key Laboratory of Molecular Targeted Drugs for Hepatobiliary and Pancreatic Tumors in Liaoning. My team has published several studies to clarify the mechanisms of liver and pancrease cancer. These projects were supported by National Natural Science Foundation in China (NSFC), Ministry of Education in China. In addition, we have published more than 150 papers in the peer-reviewed professional journals, such as Cancer Letter, Pancreas, International Journal of Surgery. I am currently mentoring 2 PhD candidates and 6 MSc candidates.      ?

1.? Pancreaticoduodenectomy    ? ? 

Pancreaticoduodenectomy (PD) is the standard operative procedure for a variety of malignant and benign diseases of the pancreas and periampullary region. With improvements in surgical technique and perioperative management, the mortality rate associated with this procedure has decreased to less than 5 %; however, the overall postoperative complication rate remains high, at 30–60 %. Postoperative pancreatic fistula (POPF), with a prevalence of 5–35 % after PD, is the most feared complication, leading to many other postoperative complications and deaths. Therefore, we performed several studies to evaluate different strategies to prevention and treatment of POPF. Our studies showed that pancreaticogastrostomy is superior to pancreaticojejunostomy. In addition, pancreaticoduodenectomy is safe for elderly patients.      ?

2.? Pancreatic neuroendocrine neoplasms    

Neuroendocrine neoplasms (NENs) are epithelial tumors with a predominant neuroendocrine differentiation, and they can develop in most organs. This fairly rare neoplasms displays a large spectrum of clinical presentations. According to the Surveillance, Epidemiology and End results (SEER) database, more than half of all NENs are gastroenteropancreatic NENs (GEP-NENs) (61%), with the highest frequency being observed in the rectum (17.7% of NENs), small intestine (17.3% of NENs) and colon (10.1% of NENs), followed by the pancreas (7.0%), stomach (6.0%), and appendix (3.1%). The annual incidence of GEP-NENs is about 3.65-4.7 cases per 100,000 people in the United States (USA). African Americans show a higher incidence than Caucasians (6.46 vs. 4.60 per 100,000). The incidence is also slightly higher in men compared with women (4.97 vs. 4.49 per 100,000). Similar rates were reported in Sweden, Norway, Spain and England. Many studies reported the epidemiology, diagnosis, pathology and management of GEP-NENs in the American and the European populations, but there is a lack of data in Asian populations, especially in China.Therefore, the objective of the present study was to perform an epidemiological study of GEP-NENs in a Chinese population. The present study might provide new clues about the development and the management of these rare tumors.    ? ?

3.? Autophagy    

Autophagy and Hippo-YAP1 signaling pathway, two processes, has been reported to regulate the pathophysiology of cancer and the sensitivity to chemotherapy. We tried to clarify mechanisms of autophagy and Hippo-YAP1 pathway. We observed that bafilomycin A1, an inhibitor of autophagy, induces caspase-independent cell death in hepatocellular carcinoma cells via targeting of autophagy and MAPK pathways. In addition, we found tumor suppressor Spred2 interaction with LC3 promotes autophagosome maturation and induces autophagy-dependent cell death.
