A.?? Personal Statement

My research interests are on the isolation, structural characterization, biological evaluation, structural optimization, and mechanistic exploration of natural products produced by insects, Ganoderma fungi, and plant resins. I have focused on compounds with potential reno-protective and neuro-protective effects inspired by traditional Chinese medicine philosophy or bionic concepts. In particular, I made great progress on nonpeptide small molecules from the insects and aromatic meroterpenoids from Ganoderma fungi, which represents pioneering work in this field. Given these achievements, I was supported by the National Science Fund for Distinguished Young Scholars in China. My future research will utilize biotic and abiotic stressors, such as light induction, to create chemical and biological diversities of metabolites from the above materials. Under the support of projects such as National Major Scientific and Technological Special Project for “Significant New Drugs Development” during the Eleventh and Twelfth Five-year Plan Periods, NSFC-Joint Foundation of Yunnan Province, National Natural Science Foundation of China, Knowledge Innovation Project of the Chinese Academy of Sciences, and West Light Foundation of the Chinese Academy of Sciences, I have published 200 papers (e.g. JASN, Org Lett, J Nat Prod) and appliedfor 67 patents. I am currently mentoring two PhD candidates and 11 MSc candidates.

1.????? Bioactive compounds from medicinal resins

Resins are secreted by many plants, and their biological effects and defensive roles in the plants have been documented. For example, Gambogic acid is a xanthonoid derived from the resin of Garcinia hanburyi, and is well known for its profound antitumor activity in clinical trials. Resina Toxicodendri (ResT) is the dried medicinal resin produced by T. vernicifluum. ResT is used as a Chinese medicine for promoting blood circulation and removing blood stasis and always occurs in prescriptions of Chinese medicine. We hypothesized that ResT might contain compounds that are beneficial for microvascular-associated diseases, such as diabetic nephropathy and renal fibrosis. With this motivation, renoprotective compounds targeting the TGF-β1/Smad signaling pathway were characterized. Our study revealed that GQ5 could be a promising drug lead for developing an effective anti-fibrotic remedy. Structural optimization of GQ5 is undergoing.

2.????? Nonpeptidal small molecules from the medicinal insects

Insects are the most diverse and abundant of all terrestrial animals. While the crucial role that insects have in the ecosystem has received great attention, studies of the chemical defense mechanisms used by insects began only in the late 1950s. Since the inception of these efforts, several structurally interesting small molecules, which are utilized as defensive substances, have been discovered. However, in contrast to efforts exploring their species diversity and richness, as well as their peptide components, much less attention has been given to non-peptidal chemical substances that insects employ to combat natural enemies or maintain their physiological activities. To an even greater extent, the potential roles that insect-derived substances might have as lead compounds in drug discovery efforts has been largely ignored. Insects are utilized as medicines or foods in some cultures of the world, especially China. The insect, Aspongopus chinensis Dallas (Pentatomidae) is an endemic food and medicinal food found mainly in southern China. The dried body of A. chinensis, is a common traditional Chinese medicine utilized to treat pain, poor digestion and kidney diseases: Yang has effects on promoting the flow of Qi, warming the stomach, relieving pain and restoring the kidney. We thus undertook an investigation on this insect and characterized >100 small molecules including novel natural product hybrids. We then assessed the biological activities of these molecules on renal protection. Understanding the mechanisms underlying the formation of hybrids in A. chinensis and compounds from the other medicinal insects is undergoing.

3.????? Structurally diverse meroterpenoids from the fungal Ganoderma

Ganoderma species are a collection of mushrooms that are mainly found in tropical regions. Several species in this genus are of medicinal value and have been used as famous traditional Chinese medicines for centuries. Amongst these important species is Ganoderma lucidum, also named G. lingzhi, which has attracted great attention for decades. In 2009, we initiated an investigation on G. lucidum, and reported the first reno-protective meroterpenoid enantiomers (lingzhiols) in 2013. We have since described a number of structurally diverse meroterpenoids from G. lucidum or the other Ganoderma species with reno-protective or neuro-protective activities. These findings not only add new facets for the compound classes in the genus Ganoderma, but also imply that there is a biosynthetic pathway responsible for generating a pool of such diverse meroterpenoids, which might arouse great interest in the biochemistry field. Thus far, the total synthesis of lingzhiols has been successfully performed. In the future, we will use fermentation and/or biotic and abiotic stress to induce Ganoderma fungi to generate structurally and biologically diverse compounds and explore their roles in the intervention of chronic disorders.
